Accumulating evidence from preclinical and clinical studies has shown that green tea catechins play a role in ameliorating the pathological characteristics in obesity, type II diabetes, cardiovascular diseases, and cancer. However, the molecular mechanisms underlying the effects of green tea catechins in each of these diseases are not completely understood. Abstract In this project, effects of PEG2 and PEG3 treatment on growth of Chlamydomonas reinhardtii (hereinafter referred to as C. reinhardtii) mutants deficient in functioning of the FA pathway is investigated.The results of this project showed that treatment of cells with PEG2 in the medium resulted in growth of the cells having an albino phenotype having yellowish autofluorescence under a microscope. A high concentration (0.5% w/v) of PEG3 caused cell growth in all strains treated with the compound. On the other hand, a low concentration of the compound (0.2% w/v) did not result in growth of the wild-type cells. Moreover, no growth was observed of the wild-type cells treated with PEG3. In the FA mutants of C. reinhardtii, cell growth was observed in the strains treated with PEG3, while the cell growth in the autotrophic strains was observed in the medium with no addition of PEG3. To investigate possible action mechanism of the compound, the possible presence of unsaturated fatty acids in the treated cells was studied. The results of this project showed that unsaturated fatty acids, such as linoleic, linolenic and arachidonic acids in the treated cells decreased depending on the concentration of PEG3 in the medium. The results of this project revealed that treatment of C. reinhardtii with either PEG2 or PEG3 caused cell growth, and it was suggested that PEG2 increases the unsaturated fatty acid content in the cells, while PEG3 has a different action mechanism. PEG2 has the growth-promoting effect probably by increasing the level of unsaturated fatty acids in the cells. In addition, it was suggested that PEG3-treated cells had an increased level of polyunsaturated fatty acids (PUFAs) and the effect of PEG3 on cell growth may be related to the presence of PUFAs in the treated cells.
